FAQ – 48-Well Platten

A 48-well plate is ideal when: (1) more than 24 conditions need to be tested, but (2) the volume of a 96-well plate (0.1–0.2 mL) is too small for downstream analysis. Example: 8 drug concentrations × 6 replicates = 48 wells—in a single plate, with enough cell material for Western blotting.

Typical: 2–5 × 10⁴ cells/well for fast-growing cell lines. For primary cells and neurons: 1–2 × 10⁴ cells/well. Maximum capacity: approximately 3–5 × 10⁵ confluent cells/well. Seed 24 hours before the experiment to allow for adhesion.

Yes — 100–200 µl of supernatant from the 48-well plate is sufficient for most ELISA kits (typical sample volume 50–100 µl). Collect the supernatant without disturbing the culture and store it at -80°C or measure it immediately.
