Applications of the 6-well plate

Primary cells

Primary cells require maximum growth surface and optimal adhesion conditions. The 6-well plate with 9.5 cm²/well is the standard format for primary cultures: hepatocytes, neurones, cardiac myocytes, PBMC, mesenchymal stem cells (MSC). TC-treated surface ensures uniform cell adhesion without additional coating.

Organoid cultivation

Organoids and 3D cell cultures require sufficient volume and surface area - the 6-well plate is the ideal format. Combination with Matrigel/Geltrex or Ultra-Low Attachment surface (available as a variant) for spheroid formation. Maximum volume of 4.5 ml/well enables sufficient extracellular matrix embedding.

Transfection & CRISPR

Standard transfection format for all common protocols (lipofectamine, PEI, calcium phosphate). 9.5 cm²/well provide sufficient cell mass for Western blot, flow cytometry and PCR analyses after transfection. CRISPR-Cas9 editing experiments benefit from the large growth area for single-cell cloning after selection.

Virus production

Lentiviral, adenoviral and AAV vector production in HEK293T cells - 6-well plate is the standard small-scale format for virus production. Sufficient volume for calcium phosphate transfection (500 µl precipitate + 2 ml medium). Non-TC variant on request for suspension cell lines.

Wound Healing Assay

6-well format for manual scratch assay and light-based wound healing with innoME's Scratch Maker. Compatible with zenCELL owl Live Cell Imager for continuous real-time monitoring of wound healing - without manual time point measurements. Coated version (Coating Plates) available for optimised cell adhesion.

Western blot preparation

Sufficient cell material for protein extraction for Western blotting: Typically 200-500 µg total protein from a confluent 6-well plate. Direct lysis in the well with RIPA buffer. Significantly more efficient than several small wells - one 6-well plate replaces approx. 6 × 12-well plates.

FAQ - 6-well plates

Typical sowing: 0.5-1 × 10⁶ cells/well for slow-growing primary cells, 2-5 × 10⁵ cells/well for fast-growing lines (HeLa, HEK293). Adjust confluence after 24-48h. The 9.5 cm² area holds up to approx. 5 × 10⁶ confluent cells (depending on cell line and size).

TC-treated (Tissue Culture treated): Vacuum plasma treatment makes the surface hydrophilic and negatively charged - for adherent cell lines. Non-TC: Untreated hydrophobic PS surface - for suspension cells or when a proprietary coating (collagen, fibronectin, Matrigel) is applied.

Yes - innoME's coated 6-well plates are directly compatible with the zenCELL owl Live Cell Imager and the Scratch Maker for Wound Healing Assays. The zenCELL owl end-to-end system (Scratch Maker + zenCELL owl + software) is specially optimised for 6-well and 24-well formats.

Recommended: 2.2 ml (minimum for complete soil coverage) to 4.5 ml (maximum without risk of overflow). Standard for most protocols: 2.5-3 ml. For transfection with calcium phosphate: 2 ml medium after addition of the precipitate.
